Die verarmte russisch-jüdische Immigrantenfamilie mit einer langen Tradition von Rabbinern und Gelehrten verkaufte den Zweijährigen nach dem Tod der Mutter für 350 Dollar an die Adoptiveltern Doris Marie und Joseph («Joe») Brodkey. Brodkey wurde nach eigenen Aussagen von seinem Adoptivvater sexuell missbraucht. Die Nachstellungen endeten erst, als der Adoptivvater 1944 starb und die Familie verarmt zurückließ. 1946 begann Brodkey, in Harvard Literatur zu studieren. Seit den fünfziger Jahren schrieb er Kurzgeschichten, die hauptsächlich in den amerikanischen Zeitschriften «The New Yorker» und «Esquire» veröffentlicht wurden. Brodkey ließ sich als freier Schriftsteller nieder und unterrichtete sporadisch Literatur und Creative Writing an der Cornell University und am City College of the City University of New York. In den sechziger Jahren lebte er zwei Jahre lang in einer homosexuellen Wohn- und Lebensgemeinschaft mit zwei Freunden zusammen; einer seiner Freunde starb später an Aids. 1978 heiratete Brodkey die Schriftstellerin Ellen Schwamm. Die Ehe, aus der eine Tochter stammt, währte bis zu seinem Tod. Sein erster Kurzgeschichtenband «First Love and Other Stories» (1954) wurde mehrfach preisgekrönt. 1991 veröffentlichte Brodkey mit großem Medienecho den Roman «Die flüchtige Seele», 1994 erschien sein zweiter und letzter Roman «Profane Freundschaft». Seine Tagebücher wurden posthum als «Die Geschichte meines Todes» (1996) veröffentlicht. 1992 unternahm Brodkey, bereits sehr geschwächt, mit seiner Frau eine Vortragsreise nach Berlin und Venedig. 1993 teilte er in einem Essay im «New Yorker» der Öffentlichkeit mit, dass er an Aids erkrankt sei. Harold Brodkey starb am 26. 1. 1996 in New York. Auszeichnungen: Prix de Rome American Academy Award (1959); Creative Arts Public Service Grant (1972); O. Henry Award (1975, 1976 und 1978).
